PacBio RS II Upgrade:  We are very pleased to announce the RSII upgrade of our PacBio instrument.  Subsequent runs indicate that we are far exceeding SMRTCell output specifications and have achieved these results with excellent samples and libraries (as output can vary depending on sample quality and organism):

o  > 50,000 reads
o  300 - 500 Mbp yield
o  4.5 - 8kb avg read length
o   > 20kb max read length

This makes our PacBio sequencing an even more versatile asset for a wide range of genomics projects. Additionally, we are carrying out R&D on using the PacBio for transcriptome sequencing, anticipating that the long reads will help identify alternative spliceforms. Other near-term applications include microbial community analysis by amplifying and sequencing the entire prokaryotic 16s rRNA gene.

*MagBead: PacBio MagBead sequencing provides more reads at longer lengths (comparative to a standard PacBio CLR library) and reduces the sample input significantly.

Table Note: The standard result estimates outlined in the table above are based on output averages of several different projects and are to be used as a guideline. Individual project results may vary from these estimates. PacBio read-lengths and read quantities per SMRTCell are highly dependent on sample quality. 

Quality Control: NCGR is committed to providing the best possible library prep and sequencing results for every sample we process. NCGR QC-tests all samples via Qubit (quantity) and Bioanalyzer and/or field inversion gel electrophoresis (quality). 

PacBio Turnaround Time: Approximately three to six weeks after receipt of signed quote, completed "NCGR Sample Form" documentation, and experimental samples of adequate sequencing quantity and quality.

XL Chemistry: NCGR offers PacBio sequencing using the new and improved XL polymerase chemistry and V3 SMRTCells resulting in longer read-lengths (averaging 6,000 bp) and an enhanced sequencing output of up to ~500 Mbp per SMRTCell.
